药物设计是随着药物化学学科的诞生相应出现的。早在20世纪20年代以前，就开始进行天然有效成分的结构改造。直到1932年，欧兰梅耶发表了将有机化学的电子等排原理和环状结构等价概念用于药物设计，首次出现具有理论性的药物分子结构的修饰工作。
