免疫球蛋白分子可变区的氨基酸序列主要在三个区域存在很大的变异性，这些区域被称为高变区(hypervariable region)，分别定位HV1,HV2,HV3，其中HV3变化最大。由于免疫球蛋白分子用这些高变环以表面互补的方式结合抗原，因此这些高变区又叫做互补决定区(complementarity-determining region,CDR)，不同重链轻链的CDR组合决定了抗体对抗原的特异性。在高变区之间的区域氨基酸序列的变化则较小，称为骨架区，轻链和重链各有四个骨架区。
