本病是一组单核-巨噬细胞（组织细胞）异常增生的疾病，较罕见，其中有Hand-Schüler-Christian病、Letter-Siwes病和嗜酸细胞肉芽肿病。因病因均未明，故定名为组织细胞增生症X。虽然各病种累及的脏器范围（骨、皮肤、淋巴结、脑垂体等）、发病年龄、临床表现各不相同。但肺都有不同程度累及，组织学改变也同样为单核-巨噬细胞异常增生和嗜酸粒细胞浸润而形成间质性肉芽肿。嗜酸细胞肉芽肿主要损害肺脏，故以肺脏病损害为本组疾病的代表加以阐述。
