相差显微镜是荷兰科学家Zernike于1935年发明的，用于观察未染色标本的显微镜。活细胞和未染色的生物标本，因细胞各部细微结构的折射率和厚度的不同，光波通过时，波长和振幅并不发生变化，仅相位发生变化（x相位差），这种相位差人眼无法观察。而相差显微镜通过改变这种相位差，并利用光的衍射和干涉现象，把相差变为振幅差来观察活细胞和未染色的标本。相差显微镜和普通显微镜的区别是：用环状光阑代替可变光阑， 用带相板的物镜代替普通物镜，并带有一个合轴用的望远镜。
