柱色谱法，又称层析法。是一种以分配平衡为机理的分配方法。色谱体系包含两个相，一个是固定相，一个是流动相。当两相相对运动时，反复多次地利用混合物中所含各组分分配平衡性质的差异，最后达到彼此分离的目的。色谱法从发明到现在已有八十多年的历史。它是纯化和分离有机或无机物的一种常用方法。其中固定相极性大于流动相的色谱为正相色谱，相反的为反相色谱。根据相似相溶原理：混合物中在固定相中溶解度大的物质后出柱，保留时间长，难被洗脱。
