外切酶即核酸外切酶（exonuclease ）。是一类能从多核苷酸链的一端开始按序催化水解3、5-磷酸二酯键，降解核苷酸的酶。其水解的最终产物是单个的核苷酸（DNA为dNMP，RNA为NMP）。按作用的特性差异可以将其分为单链的核酸外切酶和双链的核酸外切酶。
