半定量是RT-PCR做基因表达分析的一种方法，其操作的方法是在野生型和突变体中用一个看家基因（通常是actin）做参照标准来观察目标基因在各自的表达情况（上调还是下调），所谓半定量的“半”是通俗的说法，即在看电泳图估计参照亮度一致（可看作是表达的细胞数一致）情况下，确定目标基因的表达；这是与更加精确的Q-PCR的相对定量和绝对定量的区分。
