分子的旋光性最早由十九世纪的Pasteur发现。他发现酒石酸的结晶有两种相对的结晶型，成溶液时会使光向相反的方向旋转，因而定出分子有左旋与右旋的不同结构。当普通光通过一个偏振的透镜或尼科尔棱镜时，一部分光就被挡住了，只有振动方向与棱镜晶轴平行的光才能通过。这种只在一个平面上振动的光称为平面偏振光，简称偏振光。偏振光的振动面在化学上习惯称为偏振面。当平面偏振光通过手性化合物溶液后，偏振面的方向就被旋转了一个角度。这种能使偏振面旋转的性能称为旋光性。
