致癌因素是指能使人群或实验动物群体中恶性肿瘤发病率显著增加的物质或因素。可分为物理性、化学性和生物性三类。物理性致癌因素主要是放射性，约占人类肿瘤病因的5-10%。生物性致癌因素主要是致瘤性病毒，如热带性恶性淋巴瘤（Burkitt 氏瘤）及鼻咽癌，可能与E-B病毒有关。生物性致癌约占肿瘤病因的5%左右，由于遗传因素引起的肿瘤不超过5%。其余约80%的肿瘤是由于化学致癌物引起的。确定致癌因素，需要经过动物致癌实验和人群流行病学调查。
