破伤风疫苗是用破伤风类毒素制成的。传统的生产工艺包括：在有利于产毒的液体培养基中培养产毒的破伤风杆菌，用过滤的方法收获毒素，经用甲醛脱毒制成类毒素，再经若干步骤进行纯化，最终灭菌。为提高免疫原性，类毒素以铝盐或钙盐吸附。吸附破伤风类毒素通过肌肉注射接种。破伤风类毒素较稳定，可暴露于20℃的环境温度达数月，并可在37℃下贮存数周，效力仍不会出现明显的下降。不过，如环境温度达到56℃，该疫苗在2小时内即可被破坏。含破伤风类毒素疫苗应贮存于+4 (2–8)℃ 如疫苗曾被冷冻，则不应使用。
