相差显微镜是荷兰科学家Zernike于1935年发明的,用于观察未染色标本的显微镜。活细胞和未染色的生物标本,因细胞各部细微结构的折射率和厚度的不同,光波通过时,波长和振幅并不发生变化,仅相位发生变化(x相位差),这种相位差人眼无法观察。而相差显微镜通过改变这种相位差,并利用光的衍射和干涉现象,把相差变为振幅差来观察活细胞和未染色的标本。相差显微镜和普通显微镜的区别是:用环状光阑代替可变光阑, 用带相板的物镜代替普通物镜,并带有一个合轴用的望远镜。
